Evaluation of the Extended Life Platelet(ELP) Bags for the Storage of Single Donor Platelets

Abstract
The authors evaluated the effect of five day storage of single donor platelet concentrates (SD-PCs) processed by COBE Spectra all collected in extended life platelet (ELP) bags, made with polyolefin without plasticizer. In vitro measurements were
performed for platelet count, pH and lactate dehydrogenase (LD) in five units of SD-PCs during five day storage. In vivo assessments were also done with corrected count increment (CCI) at 18-24 hours after transfusion in 19 patients with
hematologic
malignancies. During storage in ELP bags, the average platelet count per unit decreased to 67% of the initial values (immediate, 1.36¡¿10E6; after five day storage, 0.84¡¿10E6 per uL) and the mean LD markedly increased to six times of the initial
values
(immediate, 71; after five day storage, 432U/L). After five days of storage, the mean pH changed to 6.85 from the initial value of 7.31 and the platelet morphology examined by transmission electron microscopy to be well preserved. There was no
significant difference in 18-24 hour CCI between the bags transfused immediately or within 1 day (12,955) and those transfused after 3-5 days storage in ELP bags (10,680).
We conclude that SD-PCs can be stored for five days in ELP bags without significant changes of pH and ultrastructure of cells.
